In the canon Runeterra timeline, Kayn and Akali appear to have heard about each other before they met, which is hinted in Zed comic, where they openly flirt with each other, Akali openly talks about Kayn's looks (abs and the shirtless thing), but "dismisses" him because of his eyes look crazy: "You're hot, but crazy." For now we don't seem to have so much information past that in the original timeline. Both of them are (was) in an order, Akali in the Kinkou Order, having Shen as her (former) master, and Kayn in the Shadow Order, having Zed as his master who he's going to be the successor of. Both Kayn and Akali live and act as assassins in Ionia, it's probable they could have some future encounters in canon.
